COMBINING TARGETED MS2 AND MS3 APPROACHES FOR THE QUANTITATION OF BILE ACIDS IN BIOLOGICAL SPECIMENS USING THE STELLAR MASS SPECTROMETER

Cholesterol-derived primary bile acids are synthesized in the liver and stored in the gall bladder's bile. When gastric emptying occurs, these acids are vital in lipid digestion and absorption. In the gastrointestinal tract, the primary bile acids are converted by the host microbiota into different secondary bile acids and microbially conjugated bile acids. Perturbations in the gut microbiome can modify the composition and concentration of the bile acid pool, and, consequently, have an impact on human health. As a result, identifying and quantifying these compounds has become a significant area of research. We present herein a highly selective targeted MS3 approach for the quantitation of bile acids and their conjugates in biological specimens using the Thermo Scientific™ Stellar™ mass spectrometer.
