13C-Metabolic Flux Analysis of Saccharomyces cerevisiae during early and late growth phases

The metabolic states of budding yeast, Saccharomyces cerevisiae, in early and late growth phases were analyzed using the 13C-metabolic flux analysis (13C-MFA). In the early growth phase, glycolysis was dominant, with minimal TCA cycle contribution, indicating a highly fermentative metabolism under normoxic condition. In the late growth phase, glucose uptake and the oxidative pentose phosphate pathway fluxes were reduced, while TCA cycle and decarboxylation reactions were inactivated. Calculating the total ATP regeneration rate revealed a transition from fermentation to respiration. Additionally, changes in the 13C-labeling of TCA cycle intermediates indicated the activation of the glyoxylate shunt in the late phase.
