Fluorinated Liquid Crystal Monomer (FLCM) Induces Kidney Dysfunction by Disrupting PPARα-mediated Fatty Acid Oxidation

This study systematically investigated the toxicological effects of DFECB, a fluorinated liquid crystal monomer. Through in vivo, in vitro, and in silico approaches, we found that exposure to human-relevant concentrations of DFECB induced renal dysfunction, characterized by interstitial inflammation, glomerular changes, and metabolic disorders. Metabolomic profiling revealed inhibited fatty acid β-oxidation as a key factor in renal impairment, correlating with PPARα downregulation. In vitro studies confirmed DFECB-induced cytotoxicity, oxidative stress, inflammation, and energy deficit in renal cells. Molecular docking simulations elucidated halogen and hydrophobic interactions between DFECB and PPARα. The study suggests DFECB acts as a disruptor of PPARα-mediated fatty acid metabolism, causing renal dysfunction, highlighting the necessity for toxicological assessment and regulation of liquid crystal monomers.
