Two-Dimensional Imaging of ¹³C-Glucose Tracing in Metabolome Analysis Using DESI-TQ-MS

This study combined DESI with TQ-MS to perform IMS for central metabolic compounds. Pregnant mice were continuously administered 13C6-glucose, and SRM was used to selectively image 13C6-glucose and its metabolite isotopomers in fetal tissues. The method successfully visualized 13C6-labeled compounds in central metabolic pathways, which are difficult to detect with MALDI imaging, providing spatial distribution data. This system extends traditional LC-ESI-MS/MS metabolomics to two-dimensional imaging without modifying SRM settings, enabling high-sensitivity spatial analysis of metabolic pathways.
