MS Imaging of Natural Moisturizing Factors in Mouse Skin.

The skin is composed of the epidermis, dermis, and subcutaneous tissue. The epidermis is subdivided into four layers, the outermost of which is the stratum corneum (SC). The SC contains sebum, intercellular lipids including ceramides, and natural moisturizing factors (NMF) including amino acids and salts. These factors are necessary for the skin to retain moisture. The present study focused on NMF and developed a method to visualize it using MALDI-MS imaging (MSI). The SC contains a significant amount of lipids, including ceramide, which has been shown to suppress ionization of NMF in conventional MALDI-MSI measurements. To address this issue, frozen sections of mouse ears were thaw-mounted on ITO glass slides and washed with chloroform, then matrices were deposited by sublimation, and MSI analyses were performed in positive ion mode (α-Cyano-4-hydroxycinnamic acid) and negative ion mode (9-Aminoacridine (9-AA)). A subsequent MSI analysis of chloroform-washed mouse ear tissue sections, the NMFs arginine, histidine, urocanic acid, and 2-pyrrolidone-5-carboxylic acid were exclusively detected within the SC. Consequently, the present study demonstrates the potential of MSI technology in detecting NMF in tissue samples while preserving the morphology of tissue sections. This method will facilitate the analysis of skin disease models, such as atopic dermatitis and ichthyosis, in the future.
