Utilizing of Antigens with Uniformed N-linked Glycans Facilitate Epitope Analysis of Glycoproteins via HDX-MS

In our research, we intended to create a general method based on HDX-MS for examining the epitopes of antibodies reacting to glycoproteins. Furthermore, this method aims to reveal the mechanisms and efficacy of neutralizing antibodies targeting multiple virus spike proteins. To counteract the heterogeneity of N-linked glycans, we focused on the action of kifunensine, a Golgi mannosidase I inhibitor. We prepared the antigen through kifunensine treatment and verified the existence of high-mannose type glycans on the antigen using mass spectrometry. We then proceeded to carry out epitope analysis using the HDX-MS system. As a result, we successfully formulated a method to produce antigens with uniform glycans and conduct antibody epitope analysis without altering existing HDX-MS systems.
