Technical improvement of plant hormone quantification from very small tissues

Plant hormones are small molecules that play crucial roles in physiological processes such as growth and environmental responses. Since their endogenous concentrations are extremely low, quantification of plant hormones has been carried out using relatively large amounts of plant materials, which are normally mixtures of whole plants or organs. Therefore, it is difficult to understand the distribution of plant hormones at the tissue or cellular levels and to detect local changes in their levels by the method. In this study, we established a technique to quantify plant hormones from very small regions within plant tissue sections prepared by Laser Micro Dissection (LMD) with higher precision. The hormones extracted from a few cryosections were quantified using a quadrupole kingdon trap mass spectrometer equipped with Nano-LC, instead of conventional LC/MS. This approach resulted in significantly higher sensitivities and mass accuracy compared to the previous methods. We will present details of the procedures as well as comparisons of data obtained with conventional and newly developed techniques.
