Noninvasive Sampling Devices Combined with Ambient Ionization Tandem Mass Spectrometry for Rapid Characterization of Metabolic Biomarkers, Ingested Medicine, and Abused Drugs in Breath

Traditional analytical methods such as GC/MS and LC/MS for characterization of the compounds in biological fluids require complex sample preparation processes, invasive sampling and lengthy analysis time thus on-site sampling and analysis are unable to be achieved. Thermal desorption-electrospray ionization tandem mass spectrometry (TD-ESI/MS/MS) operates at room temperature and requires little or no sample preparation therefore it has been successfully used to characterize chemical compounds in various biological fluids in the fields including emergency medicine, food safety, antiterrorism, anti-drug, forensic sciences, environmental sciences, and pharmaceuticals. In this study, TD-ESI/MS/MS combined with noninvasive breath sampling devices including facial mask, spirometer, and breathalyzer were developed to characterize metabolic biomarkers, ingested medicine, and abused drugs in the breath. The breath samples on the layer of facial mask, filter of spirometer and breathalyzer's mouth piece were collected with cotton swabs and analyzed using TD-ESI/MS/MS, respectively. The approach successfully detected the ion signals of trace metabolites and ingested medicines such as histamines, amino acids, caffeine, paraxanthine, acetaminophen, bupropion, plaquenil, mocalm, diclofenac, famotidine, carvedilol, and more. As a miniaturized mass analyzer is available, this technology will be used for simultaneously performing breath alcohol and drug screening tests during police check by road-side.
