Comprehensive metabolomic profiling method for disease by non-target LC/IM/MS/MS

We developed a highly sensitive, selective analytical method for steroids in serum using untargeted liquid chromatography-ion mobility-mass spectrometry (LC/IM/MS). Traditionally, immunoassays and LC/MS/MS have been used for steroid analysis, although these methods are applicable to single compound or target compounds. Our approach enables both targeted steroid quantification and the simultaneous comprehensive detection of metabolites, including phosphatidylcholine and phosphatidylethanolamine. In this study, a combined LC/IM/MS and MS/MS approach was investigated to improve the detection sensitivity of several metabolites. MS/MS performing CID over a wide m/z range enhanced detection sensitivity and allowed for non-targeted analysis. This approach provides extensive information in metabolomics. It could be applied in the future for statistical and machine learning analysis for disease research and diagnosis.
