PGE2 Is Localized Outside the COX2 Positive Implantation Site during Pregnancy

Prostaglandins (PGs) are one of the bioactive metabolites involved in various inflammatory reactions. Among them, PGE2 is known to play an important role in enhancement of vascular permeability in uterus endometrium during implantation. Since COX2-PGE2 axis plays a critical role for determination of uterus implantation site, visualization of COX2 localization by immunohistochemistry has been widely utilized to study mechanism of complex embryo-uterus endarterium interaction. However, it is not clear whether the localization of the COX2 coincides with that of PGE2, therefore, it is necessary to determine the localization of the metabolite PGE2.

The samples were analyzed by DESI-MSI at 40µm pixel size using a XevoTM TQ-XS triple quadrupole mass spectrometer. PGE2/D2 and COX2 was visualized by MS imaging and immunohistochemical (IHC) respectively in serial sections of the uterus.

In this study we found that the localization patterns of COX2 and PGE2 were different, with the latter being more widely distributed at the site of implantation. Imaging mass spectrometry (IMS) allowed us to visualize trace amount of signalling metabolites and showed that PGE2 was distributed within the stromal cells where COX2 was not localized.
