Surface-assisted laser desorption ionization-mass spectrometry (SALDI-MS) has been applied in various scientific fields. Carbon nanomaterial has attracted more attention as a SALDI material because of its excellent capability for LDI process. In this study, a graphite sheet with randomly grooved nanostructure surface consisting of sp2-conjugated atomic carbon was developed as a SALDI material. The results indicated that the graphite sheet possessed an excellent UV absorption capability and showed higher ionization efficiency. Then, graphite sheet-assisted LDI-MS successfully detected amino acids and peptides in positive and negative modes with higher ion intensities and less noise than conventional organic matrix-assisted LDI-MS. Furthermore, the analysis of polyethylene glycol polymer indicated that the detectable mass range of graphite sheet-assisted LDI-MS could be up to the molecular weight of ~2000 m/z. Finally, the graphite sheet-assisted LDI-MS with simpler preparation steps provided a greater number of peaks from soy sauce than those by MALDI-MS in positive mode and showed great linear correlation between the intensity ratio and the concentration of spiked glucose in soy sauce. Thus, these results demonstrated that graphite sheet-assisted LDI-MS possesses the potential capability of small organic compound analysis in various scientific fields.
