13C-metabolic flux analysis of lipid metabolism was developed using data-independent acquisition (DIA)-based lipidomics. Fragment ions corresponding to glycerol backbones and polar heads was investigated by LC-QTOF/MS. 13C-labeling data of the fragment ions of glycerolipids and phospholipids in budding yeast was analyzed. Flux distribution of a glycerolipid metabolic pathway was successfully estimated by the time-series 13C-labeling data.
