MALDI glycotypying法による偽結核菌O抗原の解析

Y. pseudotuberculosis is a Gram-negative bacterium belonging to the Enterobacteriaceae family. In humans, infection with Y. pseudotuberculosis typically leads to gastroenteritis symptom such as diarrhea and abdominal pain. The O-antigen, a glycan located on the surface of Gram-negative bacteria, exhibits diverse structures within the same species. Determining these structures is crucial for epidemiological studies to identify infection sources during outbreaks. The conventional method for O-antigen identification involve PCR or immunologic tests, but these approaches require specific antibodies or PCR probes for each antigen, making them complex and time-consuming. Matrix-Assisted Laser Desorption/Ionization Mass Spectrometry (MALDI-MS) has been widely adopted worldwide as a rapid method for bacterial species identification. However, the utilization of MALDI-MS is currently restricted to species identification and is not employed for antigen identification. In our previous study, we developed MALDI glycotypying to determine the molecular mass of the repeating units and the monosaccharide composition of the E. coli O-antigens. In this study, we detected signal patterns derived from the O-antigens present in the four strains of Y. pseudotuberculosis, enabling to the discrimination and identification of the O-antigens.
