Imaging mass spectrometry has been developed as a method to visualize the localization of bioactive molecules. The main hurdle is the low abundance of such signaling molecules. Prostaglandins, for example, are present in concentrations as low as picograms per milligram tissue weight, requiring high sensitivity.
Triple quadrupole instruments capable of imaging can target these low abundant molecules by monitoring the generation of specific fragment ions and integrating the signal over tens of milliseconds. With these features, DESI-TQ-MS visualized the localization of monoamines in the brain with high spatial resolution without derivatization or matrix application. In addition, the previously difficult visualization of prostaglandins in the brain was achieved.
