涙液プロテオミクスにおけるDIA法の有効性評価

We have been analyzing changes in the protein of tear fluid using data dependent acquisition (DDA) proteomics to investigate the effect of contact lens wear on ocular surface. In this study, the efficacy of data independent acquisition (DIA) proteomics for tear fluid was evaluated. The tear samples collected from four subjects were analyzed using both DDA and DIA methods. The number of proteins identified using DIA method was about 3.7 times higher than DDA method (p < 0.01). Over 1,000 new proteins not detected by DDA method were detected by DIA method, including kallikrein and angiotensin-converting enzyme, which have been reported as biomarkers for glaucoma. Taken together, these results suggest that DIA method is effective for search of new biomarkers in tear fluid.
