HILIC/MS/MSによるプロテオミクス分離基盤の構築

Hydrophilic Interaction Chromatography (HILIC) coupled with liquid chromatography/tandem mass spectrometry (LC/MS/MS) is a promising technique for bottom-up proteomics. HILIC utilizes a high organic content solvent for the mobile phase, improving the MS sensitivity and throughput. Despite the potential, challenges in peptide separation robustness hinder practical application. To address this, a Two-step dissolution method was developed, optimizing peptide solubility with 25% ACN followed by adjusting to a final 95% ACN concentration. This approach significantly enhanced robustness, leading to the identification of over 1,200 proteins—20% more than conventional methods. The Two-step dissolution method demonstrated high reproducibility, suggesting its potential to improve the robustness of HILIC/MS/MS-based bottom-up proteomics.
