Steroid hormones derived from cholesterol of a precursor are biosynthesized in the ovaries, gonads, and adrenal cortex. The localization of steroids in biological tissues are required for the assessment of physiological changes associated with disease risk and the diagnostic of various diseases simultaneously by matrix assisted laser desorption (MALDI) mass spectrometry imaging. However, it is difficult to detect due to the fact that the molecules are not easily protonated/deprotonated in MALDI. Chemical derivatization method has been used as a strategy for improving the low sensitive detection of poorly ionizable molecules by MALDI. In this study, we detect derivatized steroids in animal adrenal glands using surface-assisted laser desorption/ionization (SALDI) mass spectrometry imaging with metal film.
