Surface assisted laser desorption/ionization mass spectrometry (SALDI-MS) is a method which a sample is adsorbed onto a porous material and ionized by laser irradiation. This study reports on ion formation mechanism in SALDI-MS using agar. The measured compounds were suspended in activated carbon and applied to agar. As a result, [M+H]+ or [M+Na]+ were detected, depending on the magnitude of the gas-phase basicity. Furthermore, Arabidopsis thaliana was measured using agar. As a result, the [M+K]+ peak of abscisic acid was locally detected. In summary, it is clear that the ionic species detected in agar-based SALDI-MS depend on the gas-phase basicity of the compound and the environment.
