5,000試料連続測定における瞬速マシンガンプロテオミクスの性能評価

Bottom-up proteomics using nanoLC/MS/MS has become widely used in clinical applications and in the elucidation of biological function, but balancing its throughput with robustness, sensitivity, and analytical depth remains a challenge. Increasing flow rate and shortening gradient time have been examined to improve throughput, but often result in lower sensitivity and lower depth of analysis than standard nanoLC/MS/MS. Here, we focused on capillary-flow LC to maintain sensitivity and developed an ultrahigh-speed LC/MS/MS that achieves 1.4 minutes per run, equivalent to 1000 samples per day. The method was shown to be remarkably robust, capable of more than 5,000 continuous measurements per week, including maintenance time.
