1細胞ナノデリバリーシステムによる細胞への薬物直接デリバリー方法の開発

In this study, we have developed a method for direct intracellular injection of drugs using a single-cell nano-delivery system. We have also developed a method for analyzing injected drugs and their metabolites in cells by single-cell mass spectrometry. A mixed reagent of amiodarone and FITC-dextran was injected into HepG2 cells under microscopic observation using a single-cell nano-delivery system. The cells injected with the drug survived for more than 24 hours, indicating that the influence of injection on the cells was low. After incubation of the cells for 24 hours, cells in which FITC-derived fluorescence was detected were collected using a single-cell sampling system. As a result of mass spectrometry analysis of these cell samples, a peak of injected amiodarone was detected at m/z 646.03. These results show that these methods can be used to inject drugs directly into cells.
