超臨界流体抽出/超臨界流体クロマトグラフィー―中真空化学イオン化による組織 切片中の脂肪酸の迅速分析

Medium vacuum chemical ionization (MVCI) mass spectrometry (MS), a novel technique developed in our laboratory since 2021, utilizes oxonium and hydroxide ions generated from the water via corona discharge for chemical ionization. A great advantage of MVCI is orders of magnitude higher detection sensitivity over electrospray ionization. MVCI reaction is carried out in approximately 100 Pa, readily connected with supercritical fluid chromatography (SFC). Fat-soluble molecules can be directly extracted by supercritical fluid extraction (SFE) from a small section of tissue or a cell placed on a frit, which can be introduced into SFC using hydraulics. The synergistic combination of high peak resolution in SFC and the elevated ionization efficiency of MVCI significantly enhances molecule identification performance.
