1.Introduction to highly accurate microbial metabolomics using ZipChip CE-MS and its application

A ZipChip CE system is a microfluidic CE device providing quick separation and sensitive detection owing to nano-electrospray ionization from underivatized and tiny amounts of analytes (e. g. amino acids). Combining the ZipChip CE system and the Orbitrap Fusion Tribrid mass spectrometer enable us to improve sensitivity and accuracy of microbial metabolomics. In this seminar, we will introduce development of a novel 13C tracer-based metabolomics and its application on a hydrogenotrophic methanogen to identify metabolic pathway. The novel method requires only 1 μL of 100 nmol/L amino acids, which corresponds to those from approximately 1 × 104-105 prokaryotic cells. Total run time, including washing steps, is 15 minutes for each sample.
