In this study, we have developed a method for selective and sensitive analysis of phospholipids constituting cell membranes at the single-cell level. In addition, optimal detergents were investigated to solubilize cell membranes and extract phospholipids. Phospholipids are assumed to play vital roles in cell membrane composition and in realizing signal transduction. Phospholipid profiles vary distinctively with cell types; particularly, specific phospholipid molecules exhibit notable increases or decreases in cancer cells. This method was applied to phospholipid analysis of cancer and normal blood cell models, and each cell-specific phosphatidylcholine peak was detected.
