Sphingolipids have various physiological functions, and act as lipid mediators. Recently, attention has focused on the distribution of sphingolipids in the tissues and being a biomarker in a clinical region. Recently, MALDI-IMS techniques were established and useful for the distribution analysis of metabolites. However, only MALDI-IMS data could not demonstrate the functional concentration of metabolite. In addition, sphingolipids in tissue slice has to be taken into account degradation of compounds at room temperature during sample preparation. Therefore, we developed a new strategy of combination with LC-MS/MS and IMS technologies for sphingolipids analysis in clinical samples, and analyzed human tissue and plasma samples.
