超臨界流体クロマトグラフィー三連四重極型質量分析によるワイドターゲット定量リピドミクス手法の開発

Multiple reaction monitoring (MRM) using triple-quadrupole MS (QqQMS) allows for the reliable monitoring of low abundance lipids because of its highly sensitive and selective performance. Due to the sample complexity, a fast-scanning MRM acquisition is required to provide enough coverage of the lipidome. Herein, we have developed a novel analytical approach for wide-targeted and quantitative analysis of lipids in different biological samples using supercritical fluid chromatography (SFC) coupled to a fast-scanning QqQMS (SFC/MS/MS) including theoretically calculated comprehensive lipid MRM libraries. Our strategy was as follows: (i) preparation of a mixture of small aliquots from each sample extract, (ii) selection of target lipids by SFC RT-scheduled MRM screening methods (150 MRM transitions × 16 runs), and (iii) targeted quantitative analysis of lipids in all samples by performing a single-run SFC RT-scheduled MRM method. Our advanced analytical method will be useful for in-depth studies on complex lipid metabolism, as well as biomarker research.
