Surface-enhanced Raman scattering spectroscopy (SERS) and surface-assisted laser desorption/ionization (SALDI) are two complimentary laser spectroscopic methods in quantitative chemical analysis. However, there is no report how the nanostructures of silver materials determine their dual-functional SERS/SALDI activity so far. In the present study, we developed a silver nanoparticle-modified glass fiber (Ag NP-GF) substrates for simultaneously enhanced SERS/SALDI substrate. The optimal amount of Ag deposition and surface morphology were for the first time determined that yield maximum signal enhancement in both SERS and SALDI. The closely-packed Ag NPs with each size of 20-50 nm and the inter-particle nanoscale gap of less than 10 nm were effective for simultaneously enhanced SERS/SALDI substrate, via plasmonic/thermal hot spots among closely packed Ag nanoparticles. In addition, we explored the ability of Ag NP-GF substrates for the quantitative SERS/SALDI-MS analysis. We believe that this contribution is theoretically and practically relevant because it demonstrated a new effective and reproducible substrate for combined SERS/SALDI analysis using glass fiber for the first time.
