オンライン超臨界流体抽出 -超臨界流体クロマトグラフィー質量分析を用いた脂溶性ビタミンの一斉分析系の構築

Fat soluble vitamins (FSV) including vitamin A, D, E, and K play an important role in regulating physiological functions in vivo. Previous studies in various animal species have demonstrated that not only deficiency of FSVs but also excess intake lead to serious diseases or pathological conditions, therefore it is important to analyze FSVs accurately. However, an ideal method for FSVs quantification has not been fully developed, since FSVs are very sensitive towards exposure to extreme pH, oxygen, light and heat. Accordingly the development of a novel analytical system that enables the accurate and simultaneous measurement of FSVs is needed for the clinical diagnosis, as well as for the underlying mechanisms of FSVs related diseases. The objective of the present study was to establish a simple and practical methodology for the quantitative analysis of FSVs based on the supercritical fluid extraction-supercritical fluid chromatography/triple-quadrupole mass spectrometry (SFE-SFC/MS/MS) which enables the online extraction, chromatographic separation, and detection under dark and oxygen-free conditions.
