MALDI-ISD Analysis of Protein Containing Disulfide Bridges

In-source decay (ISD) coupled with matrix-assisted laser desorption/ionization (MALDI) is useful for amino acid sequencing of peptides and proteins. However, when a protein contains disulfide bonds, ISD peaks decrease in intensity due to the influence of disulfide bond, and it is difficult to obtain sequence information. It has been reported that disulfide bonds of protein are cleaved by hydrogen-donating matrix 1,5-DAN. In this study, the relationship between ISD peaks and disulfide bonds was considered from resulted spectra. In addition, in order to obtain further sequence information, we performed a reduction treatment of disulfide bonds using dithiothreitol (DTT). When the reduction treatment was performed with DTT, the increase in the intensity of the ISD peaks was observed.
