A Comprehensive Normalizing Method for Endogenous Metabolites by Direct nanoESI – Mass Spectrometry
Direct nanoelectrospray ionization (nanoESI) – mass spectrometry is extremely useful for a single cell analysis to detect trace amount of metabolites. However, various concomitant constituents in a cell cause matrix effects upon ionization and it hinders an accurate analysis due to fluctuations in peak intensities. The extracts of algal cells cultivated with a stable isotope labeled medium were used as an internal standard mixture and it was found useful to lessen the fluctuation. This internal standard method was applied to elucidate the influence of inorganic ions such as Na+, K+, Li+, Cl-, and PO43-, concomitantly present in a cell to the ionization of cellular endogenous metabolites. There found a trend between ionization tendency and matrix effects in their ionization.
