質量分析を用いたアセチル化に伴うヌクレオソームの構造変化の解析

Post-translational modifications (PTMs) of histone tails in nucleosome core particle (NCP), such as histone acetylation, play crucial roles in regulating gene expression. In this study, to understand the regulation mechanism caused by the structural change of NCP by acetylation, two NCPs (NCP with Widom 601 DNA and α-satellite DNA) were reconstituted in vitro, acetylated with histone acetyltransferase, and characterized using native mass spectrometry. In the case of NCP with 601 DNA, artificially prepared tight-wrapping NCP, no change was detected in the mass spectra with/without acetylation. On the other hand, the peaks of dissociated DNA were significantly observed in the mass spectra of the acetylated NCP with α-satellite DNA in 1 M NH4OAc solutions. It was suggested that structural stability in high salt concentrations of the acetylated NCP with α-satellite DNA was lower than that of the unmodified NCP. Additionally, it will be discussed the relationship between the structural stability of acetylated NCP and epigenetic regulation.
