シアル酸の誘導体化によるNASHモデルマウス肝臓切片上のN結合型糖鎖のMALDIイメージング質量分析

Glycans play important roles in the physiological process and are drawing attention as biomarkers. Glycans can be detected by mass spectrometer, especially recently matrix-assisted laser desorption/ionization-imaging mass spectrometry (MALDI-IMS) has enabled the visualization of the distribution of glycans on tissues. However, MALDI-IMS of glycans with sialic acids (sialo-glycans) is still challenges because of the weakness of the detection signal due to the carboxyl group of the sialic acids. In this study, to detect sialo-glycans with high sensitivity by MALDI-IMS, we focused on derivatization of sialic acids. We investigated the reaction condition of amidation and developed a sialic acid derivatization by benzylamidation which is more sensitive than conventional method of amidation. Furthermore, we transferred this method onto formalin-fixed paraffin-embedded (FFPE) liver tissue of normal and non-alcoholic steatohepatitis (NASH) model mouse for MALDI-IMS.
