生命科学分野におけるICP-MSの応用:メタロミクス

Metallomics is a newly coined a scientific term, and is defined as the study of metallome, i.e., the "comprehensive analysis of the entirety of metal and metalloid species within a cell or tissue type". Because targets of metallomics are a trace amount of metals/metalloids in biological samples, metallomics always requires a high sensitive analytical tool. Nowadays, ICP-MS is the most frequently used technique for metallomics. Although ICP-MS is also used for speciation hyphenated with HPLC, ICP-MS can provide only elemental information of metal-containing bionolecures, the identification by LC-ICP-MS is effective only when the retention times of samples could be matched with those of certified or authentic species (standard compounds). In other words, if the authentic compound of an unknown metabolite is not available, the unknown metabolite would not be identifiable by LC-ICP-MS. To overcome this disadvantage, a complementary use of ICP-MS and ESI-MS hyphenated with HPLC is often used to explore metal/metalloid-containing metabolites. In this lecture, I intend to talk about the following topics for selenium (Se) which have been revealed by LC-ICP-MS and LC-ESI-MS; (1) Se metabolism for its urinary excretion, (2) Molecular mechanisms of Se methylation for its excretion, and (3) Identification of a novel cellular selenometabolite.
