Morpholino Antisense Oligonucleotides Sequencing Using a Matrix-Assisted Laser-Desorption/Ionization Digital-Ion-Trap Mass Spectrometer (MALDI-DIT-MS)

In recent years, the practical application of oligonucleotide therapeutics has rapidly progressed, and a simpler and more rapid analytical method is desired. We have previously reported antisense single-stranded DNA/RNA analysis and siRNA analysis using matrix-assisted laser-desorption/ionization digital-ion-trap mass spectrometer (MALDI-DIT-MS) (MSSJ, 2022, 3P-14; MSSJ, 2023, 2P-44). In this study, we report the antisense morpholino oligonucleotides analysis using MALDI-DIT-MS. Morpholino oligonucleotides are known to be difficult to fragment using collision-induced dissociation (CID). Last year, sequencing of morpholino oligonucleotides using electron-capture dissociation (ECD) combined with LC-MS was reported (Anal. Chem. 2023, 95, 16352-16358). Herein, we introduce morpholino oligonucleotides sequencing using MALDI-DIT-MS, which enables simpler and faster sequencing.
