Integrated LC-HRMS/MS Analysis of Lipidome and Other Metabolome and Application to Crohn's Disease Susceptibility Gene Knockout Analysis

The progress of lipidomics using LC-HRMS/MS is remarkable, and especially lipidomics analysis using non-target equipment is becoming mainstream. Furthermore, recently methods have been developed for extraction of both lipids and other metabolome at once, mixing the samples, and conducting an integrated analysis of the lipidome and other metabolomes. Here, the lipid one-phase extraction method that we have developed CMEA method so far is capable of recovering lipids similar to the existing Bligh and Dyer method and Folch method, but compared to these methods, it is more excellent in lysolipid recovery. Furthermore hydrophilic metabolomes can also be extracted. Focusing on the extraction characteristics of a wide range of molecules from lipid-soluble to water-soluble, we have developed a new simultaneous lipidome and other metabolome analysis method (Hydro-Lipo (HL) Metabolomics) using CMEA extracted samples.
Using this analysis method, we analyzed the ileal luminal contents and feces of Crohn's disease (CD) susceptibility gene knockout mice and WT mice in order to reveal a mechanism of IBD.
