International Comparison on Quantification of Monoclonal Antibody: Amino Acid Analysis and UV Absorption Measurement

Monoclonal antibodies are widely used in industry as biopharmaceuticals and clinical testing reagents. The concentration of antibodies is the most essential of their properties, and it is important to determine the precise and reliable concentration. To assess the measurement capabilities in the metrology institutes of member countries of the Meter Convention. CCQM-PAWG (Consultative Committee for Amount of Substance, Protein Analysis Working Group) conducted an international comparison of antibody concentration measurements with the participation of metrology institutes from various countries. We participated in this study for comparison of monoclonal antibody quantification by amino acid analysis and UV absorption. In this study, the anti-SARS-CoV-2 spike protein monoclonal antibody was used as the analyte. Quantification by amino acid analysis and by UV measurement were performed with participating 12 and 4 institutions, respectively.
