Development of a Targeted Lipidmics Method for EVs Using UHPLC-MS/MS

It has been suggested that proteins, nucleic acids and lipid species contained in extracellular vesicles (EVs) differ depending on the onset and progression of disease. However, current lipidomics analysis is insufficiently precise, and the establishment of an accurate quantitative analysis method is required. We therefore developed an EVs targeted lipidomics method using UHPLC-MS/MS. Then, EVs targeted lipidmics analysis by UHPLC-MS/MS successfully detected lipid molecular species including ceramides and phospholipids, lysophospholipids, ceramides, hexosylceramides, cholesterylesthers, and triglyceride. Our targeted lipidmics may utilize to the biomarker discovery for plasma derived EVs and demonstrate EVs’ related disease mechanism.
