Charge heterogeneity, purity and molecular mass analysis of therapeutic monoclonal antibodies using CESI-MS system

Immunoglobulin gammas (IgGs) are the most common molecules used for generation of therapeutic monoclonal antibodies (mAbs). mAbs are heterogeneous mixtures and comprehensive characterization of these molecules is important to their development. Accurate molecular mass analysis can be performed using LC-MS. However that of charge heterogeneity and purity is difficult by LC-MS. On the other hand, capillary electrophoresis (CE) is powerful tool for charge heterogeneity and purity, but it cannot detect accurate molecular mass. We performed charge heterogeneity, purity and accurate molecular mass analysis using the system in which CE combine with mass spectrometry (CESI-MS) through sheath less sprayer. Some clipped IgG and charge variants can be detected using the CESI-MS system.
