LC/MS analysis of various cationic compounds with using new polymer-based HILIC column having carboxyl group

Various hydrophylic substances having cationic groups, for example, choline and acetylcholine exist in living organisms. It is very important to analyze these compounds to research several life phenomena and diseases. Typically, reverse-phase chromatography with ion-pair regent or after derivatization is used for analyzing these compounds. In order to simplify the analysis, a new LC/MS method was studied. A new HILIC column, Shodex® HILICpak® VC-50 2D, with polyvinyl alcohol base packing material modified with carboxyl groups, was used for the LC. Aqueous solution including formic acid / acetonitrile was used as eluent, and ESI-MS was used for detection. It was confirmed this method is very useful for the analysis of choline, acetylcholine, amine neurotransmitters, amino acids, amino sugars, metformin, and salacinol related compounds. It was also confirmed this column is applicable for the analysis of aminoglycosides under alkaline condition. Our described LC/MS method should be more facile with higher selectivity than the previous method.
