Simplified synthesis of stable isotope-labeled fatty acids using H218O and their application to highly accurate simultaneous analysis

We simply synthesized oxygen stable isotope labeled fatty acids simultaneously by replacing ordinary oxygen atom (16O) with 18O in the presence of H218O and attempted to apply them to mass spectrometry. The peak area ratios of unreacted fatty acids after synthesis were not detected or less than 10% in all molecular species. When calibration curves (concentration range: 8 to 1,000 ng/mL) were prepared by adding 200 ng/mL of 18O2-labeled fatty acids of the corresponding molecular species to standard solution, the coefficient of determination of all fatty acids were 0.996 or more, indicating excellent quantitative performance. Therefore, this method is considered to be very useful for simultaneous and highly accurate analysis of various fatty acids.
