Establishment of convenient sample preparation strategy with high sensitivity and excellent reproducibility in mass spectrometry imaging

Matrix-assisted laser desorption/ionization (MALDI)-mass spectrometry imaging (MSI) enables to visualize the distribution of several molecules without any labelling. MALDI-MSI is now widely used for in situ metabolomic imaging of endogenous or exogenous moleclues. In MALDI-MSI experiments, the choice of matrix deposition method is crucial for achieving successful MSI results with high sensitivity and high spatial resolution. Generally, spray-coating and sublimation methods are used for matrix application. More sensitive detection of analytes can be achieved by spray-coating method compared with sublimation, but the reproducibility is low. In this study, we established a novel sample preparation method of 9-aminoacridine (9-AA) sublimation coupled with recrystallization. Our proposed sublimation coupled with recrystallization method provided an excellent reproducibility about detection ability of each metabolites.
