Facile Identification of Positions of the Double Bonds in Unsaturated Fatty Acids by On-plate Oxidation in MALDI-MS

It has been known that there are several kinds of peptides or proteins which contain lipids linked to the side chain of some amino acid (Cys or Ser), or N- or C-terminus. For case of unsaturated lipid being linked to them, the positions of unsaturated bonds should be determined by chemical methods such as ozonolysis or charge-remote fragmentation with high-energy collision-induced dissociation in MS/MS. In our previous work, we found a rapid way using MALDI-MS to identify the position of unsaturated structure in the lipid moiety of Wnt protein. Recently, Zhou et al. reported a method, which allows the facile identification of the double bonds present in glycolipid, free fatty acid, and glycerophospholipid by oxidative cleavage under ambient air. In this study, we examined our previous method and found it useful for facile identification of unsaturated double bonds in fatty acids.
