Analytical Method for Identification of Sildenafil Citrate and Its Derivatives in Traditional Medicine Enhancer Stamina by LC-MS/MS QTOF

ABSTRACT
According to Indonesian's regulation, traditional medicine should have natural ingredients without drug chemical mixing. The use of traditional medicine containing drug chemicals are harmful to health because there were no clear dose in use. Among others, sildenafil citrate, vardenafil, tadalafil and its derivatives were often found in Indonesian traditional medicine (jamu) enhancer stamina. In order to support quality control of traditional medicine, Research Center for Drug and Food of National Agency of Drug and Food Control (NADFC) of the Republic of Indonesia had an analytical method for identification of sildenafil citrate and its derivatives by LC-MS/MS QTOF. The analysis conditions were as follows column: 2.2 μm C18 120 Å 2.1 x 100 mm; mobile phase: 90% methanol with 5 mM ammonium acetate; TOF mode: negative; source: ESI; End offset plate: 500 V; Capillary: 2500 V; Nebulizer: 2 Bar; Dry gas: 8 L/min; Dry temperature: 200 °C; MS/MS: bbCID; Is CID Energy: 0 eV; Collision Energy: 25 eV and Acquisition time factor: 1 s. In conclusion, having the optimum analysis conditions above, the method can be used for identification of sildenafil citrate in traditional medicine enhancer stamina.
