13C-metabolic flux shift analysis of rapid metabolic response of MCF7 cells against 2-deoxyglucose treatment

13C-metabolic flux shit analysis (13C-MFSA) was developed to determine rapid metabolic responses against a metabolic inhibitor treatment. MCF7 (human breast cancer) cells were cultured in DMEM containing [1-13C]glucose, then treated with 5mM of 2-deoxyglucose (2DG), an inhibitor of phospho-glucoisomerase (PGI). Time course analysis of 13C-labeling patterns of intracellular metabolites revealed that a ratio of M+1 isotopomers of 3-phoshoglycerate was decreased in 30 min after the 2DG treatment. The computer simulation analysis suggested that the results reflect a shift of metabolic flux from the glycolysis to the oxidative pentose phosphate pathway.
