Influence of solubilizing agents on plasma concentration measurement using LC-MS/MS

At the early stages of drug development, the solubilizing agents are used to dissolve poorly soluble compounds with dosing solution. Depending on the route of administration, there is a possibility that co-eluting solubilizing agents affect the ionization of the target compounds in LC-MS/MS analysis. Ion suppression of the target compounds was determined in the plasma with addition of various solubilizing agents. Polyethylene glycol 400, tween 80 and solutol HS15 affected the ionization of the target compounds. The result suggested the possibility of underestimating the actual plasma concentration by some solubilizing agents.
