Sensitive and structure-informative analysis of peptides, glycopeptides and glycans by MALDI-MS

Mass spectrometry has become an indispensable tool for analyzing proteins and their post-translational modifications including glycosylation. However, mass spectrometric analysis of glycosylation still remains challenging task due to the low ionization efficiency, labile natur and complicated branched structures involving various linkage isomers. In this presentation, I will summarize my recent research activities related to sensitive and structure-informative glycosylation analysis by MALDI-MS. The contents are as follows:
1. Labelings for enhancing ion yields of glycans and glycopeptides
2. Negative-ion fragmentation for elucidating the branched structure of N-glycans
3. Derivatization for the stabilization and linkage isomer discrimination of sialic acid residues -SALSA-
