Usefulness of the monolithic short column in high-throughput analysis using LC-MS/MS

We developed a monolithic silica short column (2.1 x 20 mm) for high-throughput analysis using LC-MS/MS. First, time required for equilibration of this column after gradient elution was examined. As a result, equilibration of the monolithic column was faster than that of particle-packed columns of the same dimension. Reproducibility of peak area in 2-min cycle analysis of 11 drugs was also better. Furthermore, this column was highly durable during repeated injections of physiological samples, and various absorptive compounds were successfully analyzed. These results demonstrate usefulness of the monolithic short column in high-throughput analysis using LC-MS/MS.
