LC-MS Imaging for simultaneous analysis of pharmacokinetics and pharmacodynamics in brain

Mass spectrometry imaging is a powerful technique to visualize of specific molecules in tissue sections. However, LC-MS/MS is not used as an imaging tool because information of spatial distribution is lost in process of extraction. Here we report a new approach for LC-MS imaging. We established a new preparation method using thermal film-based laser microdissection system. This system enables to capture target tissues with size of 5 to 150 µm in diameter and arrange them on the thermal film at regular interval so that each tissue can be separately extracted keeping spatial information. By combining this system to LC-MS/MS, we acquired a heat map of abundance of target molecules in mouse hippocampus using epilepsy model with pilocarpine. It is useful to investigate distribution of drugs or chemicals in tissue samples as a single cell size.
