Imaging mass spectrometry for pharmacokinetic profile of ubiquinol

We analyzed the localization of ubiquinol (reduced-coenzyme Q10; Re-CoQ10) in the sections of mice brain by matrix assisted laser desorption/ionization (MALDI)-Fourier transform (FT)-ion cycrotron resonance (ICR) imaging mass spectrometry (IMS) to evaluate the effect of dietary Re-CoQ10 composition in mice brain. Mice were divided into two groups and fed either Re-CoQ10 by oral administration for 14 days or not. First, we investigated Re-CoQ10 content in the brain by liquid chromatography (LC)-MS, quantitatively. IMS could visualize Re-CoQ10 at a resolution of 100μm in mice brain, and showed increased Re-CoQ10 from at olfactory bulb, striatum, corpus callosum subthantia nigra, oriens layer, midbrain, cerebellum and brain stem in mouse brain, and these part related with function of movement, memory and vital region. Thus, we prove the specific localization of Re-CoQ10 and relation with brain function.
