Development of a Quantitative Method to Selectively Analyze C-terminal Amino Acids of Peptide Mixtures

Here we aimed to develop a method to selectively quantitate C-terminal amino acids of peptide mixtures. The hydrolysis using carboxypeptidase in H218O successfully separated the non-18O-labelled C-terminal amino acids and 18O-labelled internal amino acid residues by LC-MS analysis. The C-terminal amino acid concentration of model peptide mixtures quantitated by this method was in good agreement with the theoretical value.
