次亜塩素酸により発生する酸化リン脂質のLC/QTOF-MSを用いた分析

In stimulated neutrophils, hypochlorous acid is known to be produced using hydrogen peroxide catalyzed by myeloperoxidase. This powerful oxidizing agent can react with a double bond in unsaturated lipids, and chlorohydrins can formed. In this study, we added reagent hypochlorous acid to standard unsaturated phosphatidylcholines and total lipids from bovine heart, and analyzed their products using liquid chromatography / quadrupole time-of-flight mass spectrometry (LC/QTOF-MS). Monochlorohydrins, their reaction intermediates, ketones or epoxides, and other oxides were detected. We are trying to reveal actual reactions of lipid oxidation as one mechanism of disposal of foreign substances by neutrophil myeloperoxidase.
